Agardh) ON CHOLESTEROL-FED GIUNEA PIGS<br/>60 pp <br/>Thesis Adviser: Mrs. Gemma C. Vinluan <br/>This is a descriptive – experimental study on the activity of Turbinaria Ornateas a hypocholesterolemic agent. It evaluate the combined effect of diet and treatment given simultaneously to induced hypercholesterolemic male guinea pigs. Extract were obtaines by maceration and reflux condensation methods. Through its preliminary screening it was found out that active constituents present in the Turbinaria ornata crude extract were saponin, tannin and volatile oil. Upon evaluation, this study further compares the effects of crude extract from but-butones Turbinaria ornate (test drug), simvastatin (Forcad 40mg) (positive control drug) and distilled water (negative control) on the total cholesterol level in 24 male guinea pigs. Twenty-four male guinea pigs (Cavia porcellus Gesner K) were randomly allocated to four (4) groups of six (6). These guinea pigs were given a normal diet of grass during a five (5)-day acclimatization period. The guneia pigs were fasted for twelve (12) hours after the acclimatization period. The following day, blood was extracted. This first extraction determines the baseline cholesterol level. A fifteen-day high diet followed. High cholesterol diet consisted of five (5) ml of hydrogenated oil and cholesterol powder (400mg/kg BW) which were fed each day by oral gavage in combination with their atherodiet consist of flour base with added milk powder, dried egg yolk, butter yeast, salt, sugar, fiber, and vitamin. The guneia pigs were the subjected to the four varying diets. The negative control (distilles water with their standard diet), atherogenesis group (atherodiet + cholesterol feeding with their standard diet), atherogenesis group (atherodiet + cholesterol feeding with their standard diet), positive control group (atherodiet + cholesterol feeding + simvastatin with their standard diet), test drug control group (atherodiet + cholesterol feeding + crude extract from Turbinaria ornate in addition to their standard diet). After this, the guneia pigs were subjected to twelve (12) hour fasting and the day after, blood extraction was performed for the second time. This served as the post-high diet cholesterol level. Differences between post-varying treatment cholesterol and post-high diet cholesterol level were analysis on interaction between diet and treatment and t-test to further test the significant difference in the treatments. Results of this study showed that the dose of T. ornata extract was effective in controlling the increase in body weights of the test animals; however, the dose of 503 mg/kg BW was not highly effective in preventing cholesterol buildup. <br/>
