Mariano Marcos State University, College of Health Sciences, Department of Pharmacy, City of Batac 2906, Ilocos Norte. March 2013. HYPOGLYCEMIC ACTIVITY AND TOXICITY ASSAY OF BREADNUT (ARTOCARPUS CAMANSI BLANCO) SEED EXTRACT. Adviser: Rhian Jaymar D. Ramil; Co-adviser: Prof. Janelyn V. Rojas.
In general, this study aimed to determined the hypoglycemic property of Breadnut (Artocarpus camansi Blanco) seed extract in sugar-induced hyperglycemia Swiss mice and its toxicity. The breadnut seeds were macerated using 80% ethanol for 48 hours. The plant sample was soaked in alcohol and then subjected to rotary evaporator at 60°C to obtain the pure extract. The pure breadnut seed extract obtained underwent confirmatory and pharmacological testing. One-way Analysis of Variance (ANOVA) and Duncan’s Multiple Range test were employed in the statistical analysis of data. The confirmatory test involved the Bate Smith and Metcalf Test, Ferric chloride Test, Froth test and Spot test. The result of these procedures indicated the presence of flavonoids in the seed extract of breadnut. In the pharmacological testing, 15 healthy Swiss mice were induced with sugar solution in 15 days. The tests animals were then grouped into three according to separate treatments. After 12 h of fasting, Glibenclamide was orally administered to the first group with the aid of a feeding needle, pure breadnut seed extract to the second group; and distilled water alone to the third group. Blood Glucose Level was noted before performing the experiment. Based on the results of the pharmacological testing, the administration of the treatments after 1 hour had no significant difference but 2 hours and 3 hours after administration registered significant difference. In the toxicity assay, Brine-Shrimp toxicity was performed by using the methodology of Golla et al., (2011). The numbers of dead nauplii in each cup were counted against a lighed background and were noted.
